Mike C replied: "Precautions: Neuroleptic malignant syndrome, tardive dyskinesia. Hypotension and syncope, especially in initiation of treatment. History of seizures. Hepatic, renal, cardiovascular disease. Lactation(breast-feeding is not recommended) and pregnancy. Hyperglycemia. Hyperprolactinemia. Suicide. Safety/efficacy in children < 18 years not established. Disruption of body temperature regulation. Antiemetic effect. If symptoms of infection occur, consider WBC count and differential count. Side effects: Dizziness, somnolence, dry mouth, dyspepsia, constipation, ALT (SGPT) increased, personality disorder, increased appetite, weight gain, akathisia, asthenia, tremor, postural hypotension. Rare cases of Priapism and leukopenia. Interactions: May enhance the effects of alcohol, centrally-acting drugs, antihypertensive agents; may antagonize the effects of levodopa and dopamine agonists. Decreased levels of olanzapine with carbamazepine, omeprazole. Inhibitors of olanzapine's metabolism: fluvoxamine, ciprofloxacin, ketoconazole. Concomitant smoking may increase olanzapine's metabolism."

Olanzapine is in a class of medications called atypical antipsychotics. It works by changing the activity of certain natural substances in the brain. Before taking olanzapine, if you or anyone in your family has or has ever had diabetes. * You should know that you may experience hyperglycemia (increases in your blood sugar) while you are taking this medication, even if you do not already have diabetes. If you have schizophrenia, you are more likely to develop diabetes than people who do not have schizophrenia, and taking olanzapine or similar medications may increase this risk. Tell your doctor immediately if you have any of the following symptoms while you are taking olanzapine: extreme thirst, frequent urination, extreme hunger, blurred vision, or weakness. It is very important to call your doctor as soon as you have any of these symptoms, because high blood sugar can cause a serious condition called ketoacidosis. Ketoacidosis may become life-threatening if it is not treated at an early stage. Symptoms of ketoacidosis include: dry mouth, nausea and vomiting, shortness of breath, breath that smells fruity, and decreased consciousness. Patients should be tested during treatment for elevated blood sugar. Additionally, persons with risk factors for diabetes, including obesity or a family history of diabetes, should have their fasting levels of blood sugar tested before starting treatment and periodically throughout treatment to detect the onset of diabetes. Any patient developing symptoms that suggest diabetes during treatment should be tested for diabetes"
